RAW cells were incubated for 1 hour with or without JQ1 or I-BET151, then stimulated for the times indicated with either 10 ug/ml poly (I:C) [C and E] or 100 ng/ml LPS [D and F]. They were then cross-linked and lysed and the chromatin was sheared by sonication. ChIP was performed on the solubilised chromatin extract protein using the BRD4 antibody, where 5 ug of antibody immunoprecipitated 2 mg of solubilised chromatin extract protein. In all panels, the enrichment of the Ifnb promoter was measured by qPCR, normalised to input.
