A. 0.5 mg of Gen2.2 cell lysate protein were immunoprecipitated with the indicated concentration of IRF5 antibody (S485D) and for the times indicated. The immunoprecipitates were subjected to SDS-PAGE and immunoblotted with an IRF5 antibody from Cell Signaling Technology. B. Gen2.2 cells were either left unstimulated or stimulated with the TLR7 agonist CL097 (1 μg/ml) for 30 min. 1 mg of cell lysate protein was incubated overnight with 2 μg/μl of IRF5 antibody (S485D, third bleed) or pre-immune IgG as a control. The immunoprecipitates and the supernatants were subjected to SDS-PAGE and immunoblotted with IRF5 antibody (Cell Signaling Technology). The results show that IRF5 can be immune-depleted almost completely from the cell lysates.
